Italian Biotech Aptadir Therapeutics Raises €40 Million in Seed Round Led by 4BIO Capital

Aptadir Therapeutics, an Italian biotech company focused on RNA-inhibitor therapeutics, has closed a €40 million seed financing round, demonstrating significant investor confidence in its approach to treating previously intractable genetic diseases.

The round was led by 4BIO Capital, with participation from a diverse consortium of European investors including Extend, Polo Nazionale di Trasferimento Tecnologico (via CDP Venture Capital), Angelini Ventures, Evotec SE, CDP Venture Capital’s Digital Transition Fund, Indaco Venture Partners, XGEN Venture, CE-Ventures, Kerna Ventures, Italian Angels for Biotech, and Club degli Investitori.

Advancing RNA-Based Therapeutics

The funding will be directed toward developing Aptadir’s DNMT-interacting RNA (DiR) platform and accelerating its lead candidate, CAP1-FMR1, which is being developed for Fragile X syndrome, a genetic disorder that causes intellectual disability and developmental delays.

Giovanni Amabile, Founder & CEO of Aptadir Therapeutics, commented on the achievement: “The size of this seed round reflects the huge potential of our DiRs technology and the quality of an authentically international scientific team. With the support of our investors, we now have the opportunity to develop a pipeline of experimental drugs that can restore the function of genes long considered irreversibly silenced.”

The company’s approach centers on targeting epigenetic modifications rather than addressing genetic sequences directly, representing a distinct strategy within the broader RNA therapeutics landscape. This methodology aims to unlock genes that have been silenced through natural biological processes, potentially opening treatment avenues for multiple genetic conditions.

Expanding the Pipeline

Beyond the immediate focus on Fragile X syndrome, Aptadir plans to leverage the newly secured capital to broaden its experimental drug pipeline. The DiR platform targets DNA methyltransferase-interacting mechanisms, which regulate gene expression at an epigenetic level. This approach could theoretically be applied to various genetic disorders where gene silencing plays a pathological role.

The company’s ability to attract such a substantial seed-stage investment reflects growing investor appetite for innovative genomic medicine approaches in Europe. The participation of both specialized biotech investors and established pharmaceutical players like Evotec SE signals recognition of the technology’s potential commercial applications.

European Biotech Momentum

The funding round underscores Italy’s emerging position within Europe’s biotechnology ecosystem. While the country has historically been overshadowed by larger biotech hubs in Switzerland, Germany, and the United Kingdom, Italian companies are increasingly attracting attention from international investors focused on novel therapeutic modalities.

Aptadir’s success in securing backing from investors across multiple European countries reflects the continent’s strengthening infrastructure for early-stage life sciences funding, particularly in specialized areas like epigenetics and RNA therapeutics. The involvement of CDP Venture Capital, Italy’s state-backed venture investment vehicle, also highlights government support for domestic biotech innovation as part of broader digital and health-tech transition initiatives.
